Sony launches ICS updates for Xperia go, Xperia U and Xperia sola

Sony has started rolling out Ice Cream Sandwich updates for three of its entry-level smartphones, including the Xperia go, Xperia U and Xperia sola. Aside from packing ?standard Android 4.0 goodies,? the new firmware also brings several Sony-specific changes such as:

  • New version of Sony media applications ? WALKMAN, Album and Movies
  • Extended standby mode, increasing standby time by up to four times depending on user-profile and data consumption
  • Resizable widgets ?for improved personalization?
  • Improved control of mobile data consumption
  • Recent Apps button for a clear overview of your applications

Moreover, after the update, the Xperia sola can be used with gloves so you can keep your fingers warm in cold weather. Pretty cool.

Unfortunately, this isn?t the over-the-air update and you?ll rather have to connect your Xperia smartphone to a PC or Mac and use Sony?s software. This page at Sony Mobile?s website has all the information needed to proceed.

Georgian grape pickers show government's challenge

(AP) ? The green grapes that Temuri Dolenjashvili and his wife snipped from the vines Sunday and emptied by the bucket into the back of their truck provide the only income for their extended family of five.

The harvest also gave a welcome day's work to an unemployed father and an elderly neighbor whose $75 monthly pension has to help feed her unemployed son, his wife and their sickly child.

Poverty and a lack of jobs are what worry Georgians most going into a tight parliamentary election on Monday that will decide the future of the pro-Western government of President Mikhail Saakashvili. For the grape pickers and others like them struggling to make ends meet on rich agricultural land and unable to sell their produce to Russia, the election offers some sense of hope.

Since coming to power nearly nine years ago, Saakashvili has transformed this former Soviet republic and put it on a path toward what Georgians hope will be eventual membership in the European Union and NATO.

The capital, Tbilisi, its streets once dark and dangerous, now shines. The stately historic facades along its main avenues have been restored to their former glory and the parks landscaped and lit.

Futuristic glass buildings have risen to house the Interior Ministry and Justice Ministry, their see-through walls intended to symbolize transparency. Among Saakashvili's greatest successes have been his creation of a modern police force and the eradication of everyday corruption.

His ambitious reforms and an inflow of Western investment have produced impressive economic growth and raised hopes among Georgians for a better life. Poverty and unemployment, however, have remained painfully high, especially in the countryside.

The official jobless rate is 16 percent, but this does not include those who sell vegetables they grow in their gardens by the side of the road or homemade wine in re-used plastic bottles.

Georgia has traditionally exported its vegetables, fruit, wine and mineral water to Russia, but this market has been closed since the two neighboring countries fought a brief war in 2008.

Just west of the Georgian capital, in the wine-producing region of Kakheti, the gleaming new buildings give way to small vineyards and fields of grazing sheep watched over by old men carrying wooden staffs. On rough gravel roads, horses pull carts piled high with stalks of dry corn.

Dolenjashvili and his wife had come from their home in the nearby town of Sagarejo to pick the last of the grapes in a vineyard that has been in his family for generations. This year they expected a total harvest of just under a ton, about half the amount of previous years because of two hail storms that had ripped up the vines.

They and the two others they had brought along to help with the harvest worked their way down the rows under a bright southern sun, cutting off bunches of grapes and dropping them into metal buckets.

All four said they planned to vote in Monday's election, but none would say whether they would support Saakashvili's party or an opposition coalition headed by billionaire businessman Bidzina Ivanishvili.

"I will vote for the one who will make our lives better and resolve the problem of no jobs," said the 53-year-old Dolenjashvili, whose nose and cheeks were a deep red from the sun.

One of his helpers was 54-year-old Givi Khirdaladze, who said that while he could make some money in the summer in the vineyards, he was unable to find any work in the winter. He lives with his wife and their 10-year-old daughter in state-owned barracks, unable to afford a home of their own.

Nadia Chiaberashvili, 70, wiped tears from her eyes as she described how she lives with her unemployed son and his family in a house with earthen floors and no heat or running water. She shook her head when asked how she planned to vote.

"She's afraid to say anything, afraid they will take away what little she has," Dolenjashvili's wife, Eka Sarukhanashvili, explained. In addition to her monthly pension of $75, Chiaberashvili's family receives about $62 in welfare benefits.

Many in Georgia have been hesitant to state which party they support, with polls showing a large percentage of voters declaring themselves undecided. Some express fears of repercussions, primarily from Saakashvili's United National Movement, which has come to dominate all branches of government.

Facing the first credible challenge to his rule, Saakashvili is under pressure from the United States and the European Union to prove his commitment to democracy by holding a free and fair election.

This election has added significance because it ushers in a new political system that will give greater powers to the parliament and prime minister. After Saakashvili's second and last term ends next year, the party that has a majority in parliament will have the right to name the prime minister, who will acquire many of the powers now held by the president.

Both the governing party and the opposition coalition Georgian Dream have reached out to rural voters by promising to pump money into agriculture, a sector long starved of investment. Ivanishvili, the opposition leader, who made his fortune in Russia, also has said he would work to restore relations with Moscow with the aim of opening up Russian markets to Georgian produce and wine.

For those who depend on their vineyards, these promises hold out hope.

After a morning of picking grapes under a hot sun, Sarukhanashvili set out lunch for the four of them in the shade of a tree. On an overturned crate, she placed cheese she had made from milk from their cow, pickled peppers from their garden, half of a chicken that they had raised, bread baked in a stone oven and a plastic bottle of their wine.

Once everyone was seated, she raised her glass and made a toast.

"I wish that after the election that everything will be better, that everyone will live better in a united Georgia," she said. "I wish that markets will open for our wine and that we will have such good harvests that I will be able to pay for my son's education."

Dream machine: California opens the road to driverless cars

Fifty years ago "The Jetsons" debuted with the promise of flying cars before the end of the 20th century. More than a decade later, flying cars are still the stuff of cartoons, but driverless cars are on their way.

On Tuesday, California Gov. Jerry Brown signed a bill that will allow autonomous vehicles like the ones developed by Google to be road-tested in the state. A licensed driver would still have to sit behind the wheel in case of emergency. With years of testing still ahead, Californians won't see a driverless Toyota Prius or Lexus merging on Highway 5 or cruising the Sonoma Valley until at least 2017.

Driverless cars use radar technology on the front of the vehicle, cameras mounted on top and artificial intelligence to move it safely through every imaginable traffic scenario.

Last year, Google clocked 300,000 miles test-driving its prototypes in Nevada, the only other state where driverless cars can be tested on the road. The lone recorded accident involved a fender-bender in which a human was driving, so the Google engineers know what they're doing.

Once driverless cars are part of the daily traffic flow, they will make long commutes more enjoyable for those who want to get a little work done on the way to the office. There won't be a need for a designated driver to take revelers home after New Year's Eve. Elderly drivers can give up their car keys without giving up independence, and if neither parent can take their high-schooler to soccer practice, the car can do it while the teenager texts safely in the passenger seat.

Still, consumer rights groups are concerned that Google will use the technology to compile even more information about its customers, and the Alliance of Automobile Manufacturers wonders what it will mean for automaker liability. To paraphrase Bette Davis: Fasten your seat belts -- it's going to be a bumpy ride.

GOP donor among 2 indicted in alleged resort scam

Mitt Romney with Tiffany and James Catledge at 2010 fundraiser for Meg Whitman, GOP candidate for California governor.

By Dan ChristensenBrowardBulldog.org

Two figures at the center of an alleged $164 million real estate scam marketed out of South Florida have been indicted by a federal grand jury in San Francisco on mail fraud and conspiracy charges.

James B. Catledge, an investment guru who has a history of being a major Republican Party donor, and Canadian businessman Derek F.C. Elliott, are accused of fraudulently soliciting $91.3 million from investors to build a resort in the Dominican Republic that never opened.

Each man faces up to 20 years in prison, and a maximum fine that is twice the value of the property involved.

The six-page indictment announced Friday is a bare-bones document that, despite the enormous sums involved, provides few details about the scope of the alleged fraud, how many people were burned or the identities of victims.

The indictment grew out of an FBI investigation that began 2 1/2 years ago during a civil racketeering lawsuit in Miami federal court filed by 230 disgruntled investors in the ill-fated EMI Sun Village Resort and Spa.

Broward Bulldog.org reported in January 2010 that court-appointed special master Thomas Scott, a Miami lawyer and former federal judge and U.S. Attorney, had found evidence that Sun Village was actually a huge ?Ponzi-style? scheme. He recommended that U.S and Canadian law enforcement investigate.

?The unassailable fact (is) that thousands of investors/owners and by extension their families in the U.S. and Canada, as well as other countries, have been financially destroyed,? Scott wrote in his 50-page report.

Indictment details
According to the indictment, Elliott was the president of various hospitality businesses in the Dominican Republic, including a pair of resorts. Catledge is a motivational speaker and the founder of several marketing companies that, among other things, sold investments in island resorts.

In 2005, the pair took out a 2005 bank loan to buy an old hotel near Santo Domingo they named the Sun Village Juan Dolio resort. They began renovations and recruited investors through Catledge?s Nevada-based sales and marketing companies Impact Inc. and Net Worth Solutions.

Sun Village marketed its Dominican properties out of an office in Doral in western Miami-Dade County. Money that flowed in was routed through an account at a Citibank branch in Tamarac, the Miami lawsuit said.

According to a statement released by prosecutors, Catledge and Elliott?s sales pitch ?failed to tell investors that the full commissions being taken on their investment were approximately 44 percent, that the renovations were underfunded, that investors? money was being used on other projects, and the returns they promised were unsupportable and could not be achieved.

The Miami special master?s report described Catledge as ?the main architect of the Ponzi scheme.?

Catledge and Elliott diverted nearly $69 million from investors to commissions, other projects and to pay ?guaranteed? interest to early investors, the indictment said.

?Real estate secrets of the wealthy?
More details are contained in related, but non-criminal charges brought in May by the Securities and Exchange Commission?against Catledge and Elliott. The principal allegation is that the men were involved in the fraudulent offering of unregistered investments for Juan Dolio and another Sun Village resort, Cofresi.

The SEC?s complaint contends the two men raised nearly $164 million selling timeshares and other ownership interests to approximately 1,200 investors between 2004 and 2009.? The complaint also stated the pair promised investors 5 to 12 percent annual returns, while also assuring that principal investments remained safe.

Catledge and Elliott used various sales materials to convince people to use their home equity and savings to invest in securities tied to the resorts, the SEC complaint said. One visual presentation was titled, ?Real Estate Secrets of the Wealthy.?

Neither the indictment nor the SEC complaint details what happened to the millions that Catledge and Elliott allegedly siphoned away from investors.

The SEC, however, said Catledge set up a trust in the South Pacific?s Cook Islands where he funneled more than $15 million in commissions that bypassed his marketing company. He and his families were the only beneficiaries.

Court papers filed in the now-closed investor litigation in Miami say the money went to pay for the lavish lifestyle and gambling debts of the resorts? developers.

The SEC complaint says that Elliott and his father, former Sunrise resident Frederick Elliott, originally purchased the Cofresi resort in 2003 and a year later needed additional funds to finish construction work. A mutual friend suggested they talk to Catledge, who could help them raise the capital they needed.

Net Worth sales associates soon began soliciting potential investors.

Both Juan Dolio and Cofresi were ultimately foreclosed. Investors were wiped out when both projects were sold at public auction in late 2009.

The Miami special master?s report blamed Elliott and his father for ?mismanagement and/or essential theft of investor monies? that ultimately delivered ?the fatal blows to the investors.?

Still, prosecutors or SEC attorneys have not accused Frederick Elliott of any wrongdoing.

The SEC?s complaint seeks disgorgement of all of Catledge and Derek Elliott?s allegedly ill-gotten gains. Also sought: fines and permanent injunctions against them and EMI Sun Village.

Big political donor
In 2008, while authorities contend the scam was in operation, Catledge contributed more than $100,000 to the Republican National Committee, John McCain?s presidential campaign and other Republican causes.

Catledge and his wife, Tiffany, also contributed to Mitt Romney?s 2008 presidential campaign before he dropped out of the race.

On May 28, 2008, Catledge and his wife attended what the Deseret News in Salt Lake City reported was a $70,000-a-couple dinner with President George W. Bush at Mitt and Ann Romney?s vacation home in Deer Valley, Utah.

In April 2010, the Catledges attended a California fundraiser for Republican gubernatorial candidate Meg Whitman. California records show Catledge gave $30,000 to Whitman?s unsuccessful bid.

Catledge has reported no contributions to federal candidates this election cycle.

Catledge?s attorney, Las Vegas celebrity lawyer David Chesnoff, was unavailable for comment. He previously said his client maintains his innocence.

?He categorically denies engaging in any criminal conduct,? Chesnoff said.

Romney?s 2012 presidential campaign did not respond to a request for comment.

Supporters of President Barack Obama and his various campaigns also have been linked to alleged wrongdoing, both criminal and civil.

Antoin "Tony" Rezko, a Chicago businessman and longtime Obama fundraiser, was convicted of fraud and bribery charges in 2008 in connection with an attempt to extort millions of dollars from businesses seeking to do business with Illinois state agencies. He was sentenced to 10 ? years in prison.

In April, the Associated Press reported that Abake Assongba, ?a major New York donor to Obama, had been accused in a civil lawsuit in Florida of defrauding a businessman and impersonating a bank official.

Despite last week?s indictment, neither Catledge nor Elliott have been arrested.

Catledge, 45, who lives in Rancho Santa Fe, California, received a summons and is due for arraignment Oct. 5, according to the U.S. Attorney?s Office in San Francisco.

No court date has been scheduled for Elliot, 42, who lives near Toronto. The spokesman declined comment when asked if prosecutors consider Elliott a fugitive.

Transform Your Ordinary Family Car into a Luxurious and Home ...

When you are traveling by using your private vehicle, one thing that you want very much is that you want to always feel comfortable and entertained when you board on the vehicle. For this reason, you will make your car as home-like as possible so that when you board on it, you can feel as if you never leave your home. Remember that a trip is sometimes so lengthy that you have to spend hours and even days in your vehicle before you reach your destination. If the interior of your vehicle is left as it is when you have just bought it, you will have to sit for hours in your vehicle and that is obviously a very boring and frustrating thing to do. Therefore, if you frequently use your vehicle for long traveling, you should consider modifying the interior of your vehicle so that it can be much more comfortable and appealing than it was before.

The best part of your car to modify is obviously its backseat. By modifying the centermost part of your car, you can transform your family car into an awesome recreational vehicle. There are many things that you can do to improve the overall condition of your car?s interior. If your car is a van or an SUV, you can combine the center and the back parts of your car to create a large and spacious passenger?s cabin. You can remove the center seats or turn them around so that they face backward. You can then put anything useful on the center of the cabin, such as a table.

In addition to modifying your car?s seats, you can also add additional entertaining components to your car. LED monitors, Blu-ray disk player, and sound system can be great additional components for your car. You can also add a small refrigerator or wine rack to make your car appear more luxurious.

Obesity promotes prostate cancer by altering gene regulation

Wednesday, September 26, 2012

Prostate cancer is one of the most common cancers in men and early treatment is usually very successful. However, like other cancers, obesity increases the risk of aggressive prostate disease. New research, published in BioMed Central's open access journal BMC Medicine, finds that the fat surrounding the prostate of overweight or obese men with prostate cancer provides a favorable environment to promote cancer growth.

Fat is a generally underrated organ. Not only is it an energy store but it secretes a wide range of growth factors, cytokines and hormones, including leptin and adiponectin, and is a major player in the immune system, which protects the body from infection and disease. But too much fat can cause these systems to go haywire and can increase risk of diabetes, cardiovascular disease and cancer.

An international team led by Prof Gema Fr?hbeck and Dr Ricardo Ribeiro analyzed fat, from around the prostate, taken from patients undergoing surgery for prostate disease. Samples were included from men with benign prostatic hyperplasia (BPH), prostate cancer (PC), and from men where their cancer was no longer confined to the prostate. The men were also classified as being either lean (BMI25).

Regardless of type of prostate disease the overweight men had different levels of gene activity in the fat surrounding their prostates compared to the lean men. This included genes which encode proteins involved in immunity and inflammation (such as LEP, which encodes the protein leptin), and cell growth and proliferation (including ANGPT1 which encodes angiopoietin 1), fat metabolism and programmed cell death.

Additionally the activity of more genes was altered between hyperplasia and prostate cancer, and between cancer and non-confined cancer, suggesting a gradual increase in dysregulation during cancer progression.

Prof Fr?hbeck explained, "Both LEP and ANGPT1 encode proteins which are thought to have roles beyond adipose tissue itself, especially because prostate cancer cells have receptors for leptin, and angiopoietin 1. Taken together with the abnormal activity levels of other genes they will ultimately foster fat mass growth, reduce immune surveillance, and promote the formation of new blood vessels, so producing a favorable environment for prostate cancer progression."

Dr Ribeiro continued, "In an increasingly obese population, understanding how fat, especially the fat surrounding the prostate, can influence the growth and severity of prostate cancer may provide an opportunity for implementing personalized lifestyle and therapeutic strategies."

How does the iPhone 5 battery life stack up?

The iPhone 5 started shipping (and hitting store shelves) on Friday, Sept. 21. According to Apple, the company has sold more than?five milli... Read more

Apple tells us that the iPhone 5 offers "even longer battery life" than any of its predecessors, but what does that really mean? Can the new smartphone keep up ?with ?? or beat ? the competition?

Laptop Mag's Mark Spoonauer conducted some thorough tests while reviewing the iPhone 5, just as he does when checking out other smartphones, to find out.

"Our test goes out and surfs 50 of the most popular websites in succession every 60 seconds," he told me,?in describing the testing procedures Laptop Mag sets up. "It's meant to mimic how people use their phones.?Every minute or so, you might take it out of your pocket and use it."

Apple claims that the iPhone 5 has enough battery power to offer "up to 8 hours of browsing on a cellular connection, up to 8 hours of talk time, and up to 10 hours of video playback time." In?Spoonauer's tests, the device "lasted 7 hours and 13 minutes."

To put that into perspective, consider that the Samsung Galaxy S III, a key?competitor, lasted for six hours and 55 minutes in Laptop Mag's tests. The iPhone 4S lasted five hours and 54 minutes, the HTC One X lasted 5 hours and 59 minutes, the Samsung?Galaxy Nexus lasted 3 hours and 40 minutes. The current champ, when it comes to battery life, is?the Motorola Droid RAZR Maxx, with a battery life of 8 hours and 25 minutes, according to Laptop Mag's tests.

"It simply has a bigger capacity battery," says Spoonauer, referring to the Motorola Droid RAZR Maxx. "At some point physics has to take over." There's only so much that Apple's clever software tweaks can do to boost the life of a rather small battery. Of course a device such as the?Motorola Droid RAZR Maxx stands a chance of outrunning the iPhone 5 thanks to sheer capacity. "It's amazing that it lasts as long as it does, given how small the battery is in comparison to the competition," Spoonauer points out.

"For the [battery] size, I think that the iPhone 5 offers fairly good battery life," he concludes before cautioning that he has seen some complaints about battery life?on Apple-centric forums.?

We put a lot of trust in Spoonauer's quantitative tests, though, and until the anecdotal evidence of poor battery performance is overwhelming, we'll maintain our faith in his assessment. This little gadget seems to last "longer on a charge than most 4G LTE phones."

Google Play hits 25 billion app downloads, holds celebratory yard sale with $0.25 games

Google Play hits 25 billion app downloads, offers select 25-cent titles this week

Google Play crossed a rather significant milestone this morning: something to the tune of 25 billion app downloads. While the accomplishment is weighty enough on its own -- especially given that the store also offers books, music and movies, which aren't included in this tally -- Google is celebrating in symbolic style with a number of apps and games for sale at just $0.25 over the next five days. Among the mix of discounted titles, you'll find publishers such as Gameloft, Electronic Arts, Rovio, Runtastic and Full Fat. Not to stop there, shoppers will also discover a curated collection of 25 must-own movies, 25 banned books, 25 albums that changed the world and 25 top-selling magazines.

With today's announcement, it was revealed that 675,000 apps and games now live in Google Play -- a healthy increase when compared to 600,000 titles and 20 billion installs just three months ago. As you might expect, Apple still claims the largest selection with 700,000 titles in its App Store, although with such a thin separation between the two, we may see Google Play eclipse its rival in short order.

Add 'Registered to vote' as a life event on Facebook

2 hrs.

If you're feeling patriotic and want your friends to know it, Facebook has just put out a small but timely new feature that lets you tell everyone when and how you registered to vote.

It's a big election year, and if you're one of the politically?minded who likes to share such things on Facebook, this new feature is a no-brainer. Just go to your Timeline and, where you would normally put?a status update?or photo, select "Life event" and pick "Registered to Vote" under the "Travel and Experiences" heading.

You'll be able to select location and year, and put in a story or link in case you want your friends to have that info as well. There's a little more info at Facebook's blog post about the feature.

If you're actually looking for an easy way to register, there are lots of sites that can help you; your best bet is to search for "register to vote" and find your state's official voting registration page, or use one of the easy services (like Rock the Vote) that finds it for you.

Giants not planning to use Cabrera in postseason

(AP) ? The San Francisco Giants have no plans to bring back suspended outfielder Melky Cabrera for the postseason if the club is still playing when he is eligible to return.

Manager Bruce Bochy said Tuesday that he and general manager Brian Sabean are meeting to finalize their 25-man roster for the best-of-five division series. Cabrera would be allowed to play in the NLCS if the Giants advance that far, but ? at this stage, at least ? they have no intention of adding him to the roster at any point in the postseason.

An announcement regarding Cabrera is expected before the NL West champions leave town after a three-game series against Arizona that began Tuesday night.

First, they would need to alert all the parties involved: Cabrera, Major League Baseball and the players union. Cabrera can start working out at instructional league in Arizona 10 days before his suspension ends, so that factor weighs into the mix.

"Right now, we'll discuss that. We'll probably have something before the end of this homestand as far as Melky," Bochy said. "We're talking about a lot of things, and that's one of them. I'll probably have an answer for you before we leave here to go down to San Diego. I think both on our side and Melky's side, you're getting down to the time period where he can start working out and doing some things. I think he needs to know and we need to know."

San Francisco's 40-man roster is full, so when Cabrera is eligible to be reinstated the Giants would have to make a roster move. They could keep Cabrera on the 40-man roster just in case.

Cabrera, the All-Star game MVP, is batting .346, which is the highest mark in the National League.

He was suspended for 50 games Aug. 15 for a positive testosterone test. On Friday, Cabrera asked to be removed from contention for the batting crown, saying he had no wish to win a tainted title.

The Giants have succeeded without Cabrera's big bat, clinching their second division crown in three seasons Saturday night against San Diego. And they are pushing ahead with the roster they have.

"Right now, we're focusing on the future and what's going to happen, the playoffs. We're not thinking about that," center fielder Angel Pagan said. "They'll make the decisions. We are the soldiers. If they bring him, we're teammates. We'll receive him with open arms. But if not, hey, we've got to respect the decision and move on, move forward."

Reliever Jeremy Affeldt has said he wants to fight with the faces who are currently in the clubhouse ? as they've done just fine in the month-plus playing without Cabrera.

"Exactly, and I'm sure that's the way the team thinks, too," Pagan said. "I respect any decision that the team makes. We have a job to do. And we have the mentality to go out there and finish the season strong and start the playoffs the right way."

Catcher Buster Posey said the Giants will accept whatever the front office decides regarding Cabrera, who is set to become a free agent after the season.

"I don't think it would bother us than just the extra questions," he said. "That would be the main thing, the extra questions."

Posey's .332 batting average was third in the NL behind Cabrera and Pittsburgh's Andrew McCutchen (.336) entering games Tuesday night. Cabrera was batting .346 with 11 home runs and 60 RBIs in his first season with San Francisco and is five hits shy of 1,000 in his big league career.

"I've been outspoken on the penalties. If he serves his suspension and pays the price, whatever it is, you guys know I spoke out and said I think there needs to be more of a deterrent, but if he serves his penalty, I guess I'm really not against it. That's their decision," Arizona manager Kirk Gibson said. "If he's caught again, I think he should be done."

First-Ever Treatment For Rare Childhood Aging Disease Shows ...

Drug Originally Developed for Cancer Proves Effective for Children with Progeria

BOSTON, Sept. 24, 2012 /PRNewswire-USNewswire/ -- Results of the first-ever clinical drug trial for children with Progeria, a rare, fatal "rapid-aging" disease, demonstrate the efficacy of a farnesyltransferase inhibitor (FTI), a drug originally developed to treat cancer. The clinical trial results, completed only six years after scientists identified the cause of Progeria, included significant improvements in weight gain, bone structure and, most importantly, the cardiovascular system, according to The Progeria Research Foundation (PRF) and Boston Children's Hospital. The study results were published today in Proceedings of the National Academy of Sciences (Epub ahead of print).

Progeria, also known as Hutchinson-Gilford Progeria Syndrome (HGPS), is a rare, fatal genetic disease characterized by an appearance of accelerated aging in children. All children with Progeria die of the same heart disease that affects millions of normal aging adults (atherosclerosis), but instead of occurring at 60 or 70 years of age, these children may suffer heart attacks and strokes as early as age 5, with the average age of death at 13 years.

"To discover that some aspects of damage to the blood vessels in Progeria can not only be slowed by the FTI called lonafarnib, but even partially reversed within just 2.5 years of treatment is a tremendous breakthrough, because cardiovascular disease is the ultimate cause of death in children with Progeria," said Leslie Gordon, M.D., Ph.D., lead author of the study, medical director for PRF, and mother of a child with Progeria. In addition, Dr. Gordon is a staff scientist at Boston Children's Hospital and Harvard Medical School, and associate professor at Hasbro Children's Hospital and Alpert Medical School of Brown University.?

Results Yield Improvements in One or More Study Measures for All Children
Twenty-eight children from sixteen countries participated in the two-and-a-half year drug trial, representing 75 percent of known Progeria cases worldwide at the time the trial began. Of those, 26 are children with the classic form of Progeria. The children traveled to Boston every four months to receive comprehensive medical testing through Boston Children's Hospital's Clinical and Translational Study Unit.

Treatment consisted of the FTI lonafarnib, supplied by Merck & Co., given to children orally, twice-a-day over the course of the study, under the supervision of principal investigator Mark Kieran, M.D., Ph.D., director of pediatric medical neuro-oncology at the Dana-Farber/Children's Hospital Cancer Center.

The research team, which included specialists at Boston Children's Hospital, Brigham & Women's Hospital and Dana-Farber Cancer Institute, evaluated the children's rate of weight gain compared to their pre-therapy rate as the primary outcome because children with Progeria experience severe failure to thrive, and have a consistent, very slow linear rate of weight gain over time. Researchers also examined arterial stiffness (a predictor of heart attack and stroke in the general population), bone density and rigidity (indicators of osteoporosis). Every child completing the study showed improvement in an ability to gain additional weight, increased flexibility of blood vessels or improved bone structure.

Results included improvement in one or more of the following areas:

  • Weight: One in three children demonstrated a greater than 50 percent increase in annual rate of weight gain or switched from weight loss to weight gain, due to increased muscle and bone mass.
  • Bone Structure: On average, skeletal rigidity (which was highly abnormal at trial initiation) improved to normal levels after FTI treatment.
  • Cardiovascular: Arterial stiffness, strongly associated with atherosclerosis in the general aging population, decreased by 35 percent. Vessel wall density also improved with treatment.

Following the 2003 discovery of the gene that causes Progeria, researchers identified FTIs as a potential drug treatment for Progeria. Children with Progeria have a genetic mutation that leads to the production of the protein progerin, which is responsible for Progeria. Progerin blocks normal cell function and part of its toxic effect on the body is caused by a molecule called a "farnesyl group," which attaches to the progerin protein. FTIs act by blocking the attachment of the farnesyl group onto progerin.

"In the early stages of planning for this clinical trial, we realized that my team's experience using FTIs to treat children with brain cancer could bring together PRF's preclinical research efforts and the expertise we needed to study the drug in children with Progeria," said Kieran, the study's senior author and associate professor of Pediatrics at Harvard Medical School. "The premise behind studying this drug was that by stopping the attachment of the farnesyl group onto progerin in children with Progeria, progerin may be inactivated, reducing some effects of the disease."

"PRF provides a model for disease research organizations, and is a good example of successful translational research, moving from gene discovery to clinical treatment at an unprecedented pace," added Dr. Kieran.

Since PRF's founding in 1999, the organization and its scientific partners have identified the genetic mutation that causes the disease, funded preclinical research and funded clinical trials. A second clinical trial, funded by the National Institutes of Health and PRF, is currently underway and more trials are expected to follow.

"The partnership between The Progeria Research Foundation, the research team and these courageous families was essential for success," said Dr. Gordon. "It required identifying children and their home doctors from around the globe, obtaining the essential pre-trial clinical information, transporting families to and from Boston, supplying translators both inside and outside of the hospital setting, and putting together a multidisciplinary clinical team to assess treatment effects. But it was all worth it, and I believe we have set in motion a blueprint for successful treatment trials for children with Progeria and for other rare diseases."

"The results of this study provide our family with excitement and hope for Megan's future," said Sandy Nighbor, mother of Megan, a 12-year-old child who participated in the clinical trial. "We're grateful to The Progeria Research Foundation and all of the doctors for their commitment to helping my daughter and all children with Progeria."

Progeria Linked to Normal Aging Process
Previous research shows that progerin is also produced in the general population and increases in the body with age. A number of studies successfully linked progerin with normal aging, including a causal link between progerin and genetic instability, specifically telomere dysfunction in the aging process. Researchers plan to continue researching the effect of FTIs, which may help scientists learn more about cardiovascular disease that affects millions, as well as the normal aging process.??

"One of the main reasons we achieved breakthrough results in this first trial is because of the tremendous supporters who provided funding, and helped get us one step closer to achieving our ultimate goal ? a cure for Progeria," said Audrey Gordon, Executive Director of PRF. "Every donation makes a difference. With continued support, we will fund research that will not only allow children with Progeria around the world to live long and healthy lives, but may also advance our understanding of the normal aging process that affects us all."

About The Progeria Research Foundation (PRF)
The Progeria Research Foundation (PRF) was established in 1999 to find the cause, treatment and cure for Progeria ? a rapid aging disease that causes children to die from heart disease or stroke at an average age of 13 years.? In the past 13 years, research conducted in partnership with PRF has identified the gene that causes Progeria and possible treatments.? PRF funded and coordinated this first-ever Progeria clinical trial.? PRF is currently funding a clinical trial in which children with Progeria receive FTI plus two additional medications to slow the progression of Progeria.? PRF continues to identify more children who can benefit from the programs and services that it provides while helping advance research towards treatment and cure.? To learn more about Progeria and what you can do to help, please visit www.progeriaresearch.org.
